我们参照English等(J ImmunolMethods 1974 5:249)方法,用不连续密度梯度离心一步法分离中性粒细胞(PMN)和淋巴细胞。试验取自15例正常献血员外周血,分离细胞并作PMN分离效果的检查。细胞分离方法:向试管加入约2ml分层液Ⅱ(比重1.115),然后再叠加2ml分层液Ⅰ(比重1.077)。取2ml肝素抗凝血(20μ/ml)加等量Hank's液稀释后,小心叠加在分层液Ⅰ上,2200rpm离心20分钟。结果在分层液Ⅰ界面上为单个核细胞层,在分层液Ⅰ、Ⅱ交界处及分层
We refer to the method of English et al. (J Immunol Methods 1974 5: 249) for separation of neutrophils (PMNs) and lymphocytes by one-step discontinuous density gradient centrifugation. The experiment was performed on peripheral blood from 15 normal blood donors and cells were isolated and examined for PMN isolation. Cell separation method: About 2 ml of stratified solution II (specific gravity 1.115) was added to the test tube, and then 2 ml of stratified solution I (specific gravity 1.077) was further added. Take 2ml heparin anticoagulant blood (20μ / ml) diluted with equal amount of Hank’s solution, carefully superimposed on the stratified fluid Ⅰ, 2200rpm for 20 minutes. The result was a mononuclear cell layer at the layer Ⅰ interface and at the junction and layer of the layer Ⅰ and Ⅱ layers
