目的分析PML-RARα融合基因疫苗诱导小鼠特异体液和细胞免疫应答情况。方法用成功构建的重组质粒(pIRES-PML-RARα-hIL-2)免疫小鼠后,分别利用ELISA法检测小鼠血清中INF-γ生成情况;LDH释放法检测小鼠脾细胞特异性CTL细胞针对APL细胞株NB4的杀伤率;用间接免疫荧光法检测血清中抗PML-RARα抗体。结果免疫后第7周时,小鼠血清中INF-γ含量、抗PML-RARα抗体及脾脏CTL细胞针对NB4的杀伤率均高于对照组。结论所构建的pIRES-PML-RARα-hIL-2重组质粒可诱导小鼠产生了特异性体液和细胞免疫应答。
Objective To analyze the specific humoral and cellular immune response induced by PML-RARα fusion gene vaccine in mice. Methods After the mice were immunized with the recombinant plasmid pIRES-PML-RARα-hIL-2, the production of INF-γ in the serum of the mice was detected by ELISA. The spleen-specific CTL cells Anti-APL cell line NB4 killing rate; indirect immunofluorescence assay serum anti-PML-RARα antibody. Results At the 7th week after immunization, the serum levels of INF-γ, anti-PML-RARα antibody and spleen CTL cells against NB4 were all higher than those of the control group. Conclusion The constructed pIRES-PML-RARα-hIL-2 recombinant plasmid can induce specific humoral and cellular immune responses in mice.
