仙丹升血颗粒治疗原发免疫性血小板减少症药效学研究

目的:评价仙丹升血颗粒对原发免疫性血小板减少症模型小鼠的药效学研究。方法:将小鼠随机分为正常对照组、模型组、低剂量组、高剂量组、强的松组,采用免疫法腹腔射外源性抗血清建立原发免疫性血小板减少症小鼠模型,分别运用仙丹升血颗粒、强的松治疗,观察各组小鼠外周血象、巨核细胞、脏器指数及脾脏病理学改变、出凝血试验。结果:仙丹升血颗粒能有效升高造模后小鼠血小板计数(P<0.01);使脾脏巨核细胞数、骨髓巨核细胞数恢复正常(P<0.05);高、低剂量组和强的松治疗组胸腺、肾上腺指数较模型组有明显升高(P<0.05),脾脏指数有明显下降(P<0.01),强的松组和大剂量组巨噬细胞和生发中心的凋亡小体数明显减少(P<0.05);大剂量组和强的松组治疗小鼠出血时间有明显缩短(P<0.01),且出血重量明显减少(P<0.01)。结论:仙丹升血颗粒对原发免疫性血小板减少症模型小鼠有明显治疗作用。 OBJECTIVE: To evaluate the pharmacodynamics of Xiantan Shengxue Granule on the model mice of primary immune thrombocytopenia. Methods: The mice were randomly divided into normal control group, model group, low dose group, high dose group and prednisone group. The mouse model of primary immune thrombocytopenia was established by using immunopotentiating antiserum. Respectively, using Xuedan Shengxue Granules, prednisone treatment, observed in each group of peripheral blood, megakaryocytes, organ index and pathological changes of the spleen, the clotting test. Results: Xiadian Shengxue Granule could effectively increase the platelet count (P <0.01) in mice after modeling, and restore the numbers of splenic megakaryocytes and the number of bone marrow megakaryocytes (P <0.05). The high and low dose groups and prednisone The thymus and adrenal index in the treatment group were significantly higher than those in the model group (P <0.05), and the spleen index was significantly decreased (P <0.01). The number of apoptotic bodies in the prednisone and high-dose macrophages and germinal centers (P <0.05). The bleeding time of mice in high dose and prednisone treatment groups was significantly shorter (P <0.01) and the weight of bleeding was significantly reduced (P <0.01). Conclusion: Xian Dan Shengxue Granule has a significant therapeutic effect on the model of primary immune thrombocytopenia in mice.
