以冬凌草转录组为研究对象,使用Microsatellite(MISA)软件进行SSR搜索,分析冬凌草中SSR位点信息,并利用Primer 3设计SSR引物,对其多态性进行初步评价。结果表明:研究共发现11 114个SSR,分布于8 873条独立基因(Unigenes)中,SSR位点出现频率为24.90%,共有55种重复基元,平均每3 517bp含1个SSR位点。SSR位点所包含的重复类型中,二核苷酸重复是主要类型,占总SSR的47.71%;其次是单碱基重复类型(35.46%)。SSR所包含的重复基元中,单核苷酸重复基元A/T和二核苷酸重复基元AG/CT是优势重复基元,分别占总SSR的34.95%、30.16%。多态性进行评价结果表明冬凌草转录组SSR类型丰富,多态性潜能高。
Using Rubescens indica transcriptome as the research object, the microsatellite (MISA) software was used to carry out SSR search, the SSR information was analyzed, and the Primer 3 SSR primer was designed to preliminarily evaluate its polymorphism. The results showed that 11 114 SSRs were found in 8 873 independent genes (Unigenes). The frequency of SSR loci was 24.90%. There were 55 repeat motifs, with an average of 1 SSR loci per 3 517 bp. Among the repeats contained in the SSR loci, dinucleotide repeats were the major type, accounting for 47.71% of the total SSRs; followed by the single base repeats (35.46%). Among the repeat motifs contained in SSR, mononucleotide repeat motif A / T and dinucleotide repeat motif AG / CT are dominant repeat motifs, accounting for 34.95% and 30.16% of the total SSR, respectively. Polymorphism of the results showed that the Rubusteen transcriptome SSR type rich, high polymorphic potential.
