目的:观察当归四逆汤对垂体后叶素(Pit)所致心肌缺血大鼠心电图及心肌酶谱的影响。方法:采用雄性Wistar大鼠腹腔注射垂体后叶素Pit(30 U/kg)造成急性心肌缺血损伤模型,观察各组大鼠注射Pit前10 min,及注射5、15、30 min后的心率、Ⅱ导联心电图变化,并检测血清乳酸脱氢酶(LDH)、磷酸肌酸激酶(CPK)活性。结果:注射Pit 5 min后,DSD高、低剂量组及阳性对照组心率均高于模型组,但差异均无统计学意义(P>0.05);注射Pit15 min、30 min后,DSD高、低剂量组及阳性对照组心率均高于模型组(P<0.01或P<0.05)。注射Pit后,各组大鼠心电图均出现不同程度的波形改变,主要表现为ST段向上偏移,T波高耸,心律失常等。DSD高、低剂量组及阳性对照组ST段、T波变异性程度均低于模型组。模型组LDH、CPK活性均高于空白对照组(P<0.01),DSD高、低剂量组及阳性对照组LDH、CPK活性均明显低于模型组,差异均有统计学意义(P<0.05或P<0.01)。结论:当归四逆对心肌缺血模型大鼠心电图ST段、T波、心率有改善作用;能降低LDH、CPK的活性,具有保护心肌细胞的作用。
Objective: To observe the effect of Danggui Sini Decoction on electrocardiogram and myocardial enzymes of myocardial ischemia rats induced by pituitary. Methods: The model of acute myocardial ischemia was induced by intraperitoneal injection of pituitrin (30 U / kg) into male Wistar rats. The heart rate of rats in each group was observed 10 min before injection and at 5, 15 and 30 min after injection , Ⅱ lead electrocardiogram changes, and serum lactate dehydrogenase (LDH), creatine phosphokinase (CPK) activity. Results: After 5 min of Pit injection, heart rate of high and low DSD group and positive control group were higher than those of model group (P> 0.05); Pit15 min, 30 min after injection, DSD was high and low The heart rate of the dose group and the positive control group were higher than that of the model group (P <0.01 or P <0.05). After injection of Pit, the electrocardiogram of rats in each group showed varying degrees of waveform changes, mainly as ST-segment upward shift, T-wave towering, arrhythmia and so on. DSD high and low dose group and positive control group ST segment, T wave variability were lower than the model group. The activities of LDH and CPK in model group were higher than those in blank control group (P <0.01). The activities of LDH and CPK in high and low dose DSD group and positive control group were significantly lower than those in model group (P <0.05 or P <0.01). Conclusion: Angelica sinensis can improve the electrocardiogram ST segment, T wave and heart rate of myocardial ischemia model rats, reduce the activity of LDH and CPK, and has the effect of protecting cardiomyocytes.
