用50—200毫升的烧杯,普通圆形滤纸,滤纸的直径大于烧杯口3厘米。根据种子大小在滤纸上打孔,留放种子用,以不漏下种子为宜。小粒种子的孔距约1厘米,大种子孔距为1.5厘米。另在滤纸中心打一小孔,用滤纸卷芯插入其中,作吸水用。烧杯内装满水或试剂,烧杯口铺滤纸,种子胚根端朝下,插入小孔中稍许,将烧杯放入恒温培养箱中发芽,定时补足水分,适时测量根长或苗长。所长苗的长度和其它直立发芽方法得到的苗长基本一致。烧杯透明,便于观察测量,不触及幼苗本身,在烧杯外也可测定种苗的长度。本方法适应各种种子发芽测定。
