目的研究10×Aβ3~10基因重组腺病毒疫苗在BALB/c鼠体内产生的炎症免疫反应。方法 14只3月龄雄性小鼠按随机数字表法分为目的基因组、空载体组、Aβ42组,接种相应疫苗。脾细胞培养,MTT法测各组脾细胞增殖率,ELISA法检测培养上清中细胞因子白细胞介素(IL)-4和干扰素(IFN)-γ水平。结果目的基因组和Aβ42组均在各自相应抗原重新刺激时表现出较高的脾细胞增殖率,即目的基因组在Aβ3~10多肽重刺激时脾细胞增殖率较高,而Aβ42组则在Aβ42重刺激时脾细胞增殖率较高;Aβ42组的脾细胞培养上清中IL-4和IFN-γ水平均高于空载体组,目的基因组培养上清中IL-4水平与Aβ42组相比差异无统计学意义,但IFN-γ水平显著低于Aβ42组(P<0.05)。结论 10×Aβ3~10基因重组腺病毒疫苗能够有效诱导Th2型免疫反应,而不引起针对Aβ42的T细胞免疫反应。
Objective To study the inflammatory response induced by 10 × Aβ3 ~ 10 recombinant adenovirus vaccine in BALB / c mice. Methods Fourteen male mice of 3 months old were randomly divided into the target group, empty vector group, Aβ42 group and inoculated with the corresponding vaccine. The proliferation of spleen cells was measured by MTT assay. The levels of cytokines interleukin-4 and interferon-γ were detected by ELISA. Results The target genome and Aβ42 group showed higher proliferation rate of spleen cells when restimulated by their respective antigens, that is, the target genome had a higher proliferation rate of spleen cells when restimulated with Aβ3-10 polypeptide, while the Aβ42 group was stimulated by Aβ42 re-stimulation The spleen cell proliferation rate was higher in Aβ42 group than that in empty vector group. The level of IL-4 in the supernatant of Aβ42 group was not statistically different from Aβ42 group Significance, but IFN-γ levels were significantly lower than the Aβ42 group (P <0.05). Conclusion The 10 × Aβ3 ~ 10 recombinant adenovirus vaccine can effectively induce Th2 immune response without inducing T cell immune response to Aβ42.
