目的:确定从盾叶薯蓣中提取、分离得到的一水溶性化合物(SY-A)的结构,并研究其对乳鼠心肌细胞缺血/再灌注(I/R)损伤的保护作用。方法:利用柱层析技术从盾叶薯蓣中分离到水溶性化合物SY-A,经1H-NMR、13C-NMR、135DEPT鉴定其结构;采用体外培养乳鼠心肌细胞,建立心肌缺血/再灌注损伤模型。比较正常对照组、模型组、SY-A各组(50、100、150 mg/L)培养液中乳酸脱氢酶(LDH)、肌酸激酶(CK)含量变化,MTT法检测细胞存活率。结果:SY-A结构为4-甲氧基-苯甲酸-2-O-葡萄糖苷;与I/R组比较,SY-A组心肌细胞LDH、CK的漏出量显著降低,细胞存活率显著提高。结论:盾叶薯蓣中水溶性成分SY-A对乳鼠心肌细胞缺血/再灌注损伤有显著的保护作用。
OBJECTIVE: To determine the structure of a water-soluble compound (SY-A) isolated and isolated from Dioscorea zingiberensis Wright and to study its protective effect on myocardial ischemia / reperfusion (I / R) injury in neonatal rats. Methods: The water-soluble compound SY-A was isolated from Dioscorea zingiberensis by column chromatography and identified by 1H-NMR, 13C-NMR and 135DEPT. Myocardial cells were cultured in vitro to establish myocardial ischemia / reperfusion Damage model. The levels of lactate dehydrogenase (LDH) and creatine kinase (CK) in the culture medium of normal control group, model group and SY-A groups (50,100,150 mg / L) were compared. The cell viability was measured by MTT assay. Results: The SY-A structure was 4-methoxy-benzoic acid-2-O-glucoside. Compared with I / R group, the leakage of LDH and CK in SY-A group was significantly decreased and the cell survival rate was significantly increased . CONCLUSION: SY-A, a water-soluble component of Dioscorea zingiberensis, has a significant protective effect on neonatal rat cardiomyocytes against ischemia / reperfusion injury.
