产黑普雷沃氏菌(Prevotella melaninogenica)属革兰阴性专性厌氧菌,是引起牙周疾病的病原之一。为了研究牙周炎患者产黑普雷沃氏菌是否存在及其分子特征,用棉签刮取患者口腔,沸水-液氮法破碎细胞,同时设计产黑普雷沃氏菌糖基转移酶基因的1对引物,利用常规PCR方法扩增产黑普雷沃氏菌糖基转移酶基因。经NCBI序列相似性比对,发现该患者感染的产黑普雷沃氏菌糖基转移酶基因核苷酸序列与P.melaninogenica ATCC 25845菌株的糖基转移酶基因(Gen Bank accession No.CP002122)同源性最高,达92%;其编码的糖基转移酶部分氨基酸序列与P.melaninogenica菌株的甘露糖糖基转移酶(Gen Bank accession No.WP_013264679)同源性最高,达98%。以糖基转移酶部分氨基酸序列构建系统进化树,结果表明,本试验获得的产黑普雷沃氏菌与其它报道的P.melaninogenica菌株同源性最高,属于同一组(group)。
Prevotella melaninogenica is a gram-negative obligate obligate anaerobic bacterium that is one of the causative agents of periodontal disease. In order to study the existence and molecular characteristics of Enterobacter cloacae in patients with periodontitis, the oral cavity of patients was swabbed with cotton swab, and the cells were broken by boiling water-liquid nitrogen method and designed to produce the gene of Herpes Simplexviridae glycosyltransferase 1 pair of primers, the gene of Herpesaceum glycosrans transferase was amplified by a conventional PCR method. The similarity of NCBI sequences revealed that the nucleotide sequence of the gene of Herpesacetolaήa glaucoma glycosyltransferase infected by this patient was identical to the glycosyltransferase gene (Gen Bank accession No.CP002122) of P.melaninogenica ATCC 25845 strain, And the highest homology was 92%. The partial amino acid sequence of glycosyltransferase encoded the highest homology with GenBank accession No.WP_013264679 (98%). The phylogenetic tree was constructed based on the partial amino acid sequence of glycosyltransferase. The results showed that the genus Herpescreas obtained in this study had the highest homology with other reported P. melaninogenica strains belonging to the same group.
