试验材料是从新铁炮百合(Lilium×formolongi)与东方百合‘索邦’(Oriental hybrid‘Sorbonne’)的杂交苗中筛选出的优良株系。以该株系组培扩繁的试管鳞茎为材料,通过石蜡切片观察从营养生长到生殖生长阶段的茎尖解剖结构;通过交互迁光试验初步确定光周期感受态及限界性诱导光周期;在不同光周期下,通过qRT-PCR技术测定分析了杂种苗不同组织和发育时期成花诱导相关基因CONSTANS(CO)和FLOWERING LOCUS T(FT)的表达。结合形态特征、解剖结构和基因表达状况,最终确定了该百合杂种苗成花诱导关键时期:在6~8片基生叶时开始进入光周期感受态,在2~4个节间时开始进入成花诱导期,限界性诱导光周期为28 d。
The test material was selected from the hybrid seedlings of Lilium × formolongi and Oriental hybrid’Sorbonne ’. The bulked test tube bulbs of this strain were used as material to observe the anatomical structure of the shoot apices from vegetative growth to reproductive growth stages by paraffin sections. The photoperiod and photoreceptor cycle were preliminarily determined by interacting light transfer test. The expression of CONSTANS (CO) and FLOWERING LOCUS T (FT) in different tissues and developmental stages of hybrid seedlings were analyzed by qRT-PCR at different photoperiod. Combined with the morphological characteristics, anatomical structure and gene expression status, the key stage of flower induction of Lily hybrid seedlings was finally confirmed: the photoperiod began to enter into the photoperiod when 6 to 8 basal leaves started, and began to be observed in 2 to 4 internodes Into the flowering induction period, limiting the induced light period of 28 d.
