目的 研究小鼠腹腔注射贝母乙素的药代动力学。方法 反相液相色谱法定量分析。血浆样品经乙醚提取后 ,柱前用2 ,4 二硝基苯肼衍生 ,分析色谱柱 :HypersilC18,流动相 :乙腈 乙酸铵 (0 .1mol·L-1,pH =5 ) (39∶6 1) ,UV375nm检测。结果 在0 .114~ 1.2 6 μg·ml-1区间 ,标准曲线线性方程为Y =0 .12 6X - 0 .0 0 13,相关系数r=0 .992 3,平均日间、日内精密度RSD分别为5 .85 %和 5 .30 %。平均相对回收率为 96 .5 %。结论 小鼠注射贝母乙素 (1.5mg·kg-1)的血药浓度 时间曲线符合一室开放模型 ,t1/ 2 为 0 .84h ,cmax为 0 .87μg·ml-1,tmax为 0 .15h。
Objective To study the pharmacokinetics of intraperitoneal injection of pemetrexin in mice. Methods Quantitative analysis by reversed-phase liquid chromatography.
