建立了高效液相色谱法同时测定抗抑郁候选新药CSPCHA028(化学名:N1-苄基-N4-[1-正丁基-2-羟基-2-(5-氯-6-甲氧基-2-萘基)乙基]哌嗪)的4个光学异构体。采用Chiralpak IC手性柱,以正己烷-异丙醇-二乙胺(75︰25︰0.1)为流动相,检测波长236 nm。4个光学异构体间的分离度均大于2.0,线性范围为5~120 g/ml,最低检测限均为1.0 g/ml。
A high performance liquid chromatography (HPLC) method for simultaneous determination of antidepressant drug CSPCHA028 (chemical name: N1-benzyl-N4- [1-n-butyl-2-hydroxy-2- - naphthyl) ethyl] piperazine). Chiralpak IC chiral column was used with n-hexane-isopropanol-diethylamine (75:25:0.1) as the mobile phase at a detection wavelength of 236 nm. The resolution of the four optical isomers was greater than 2.0 with a linear range of 5-120 g / ml and the lowest detection limit was 1.0 g / ml.
