目的 探讨中药咳喘落对哮喘豚鼠可溶性细胞间粘附分子 -1(sICAM -1)、嗜酸性粒细胞及其脱颗粒产物嗜酸性粒细胞阳离子蛋白 (ECP)等的影响 ,阐明咳喘落治疗哮喘抗气道变应性炎症的作用机制。方法 采用荧光酶标记法、双抗体夹心ELISA法和密度梯度细胞分离法分别测定了哮喘豚鼠血浆中ECP ,sICAM -1的含量和肺泡灌洗液中嗜酸性粒细胞分类计数。结果 哮喘豚鼠血浆中ECP ,sICAM -1的含量以及肺泡灌洗液中细胞总数和低密度嗜酸性粒细胞数均显著高于正常组 (P <0 .0 1) ,经咳喘落和地塞米松治疗后显著下降 (P <0 .0 1) ,接近正常水平 (P >0 .0 5 )。结论 咳喘落通过调节白细胞介素 -5的水平 ,抑制嗜酸性粒细胞在气道内募集及其毒性颗粒蛋白的释放 ,从而发挥它抗气道变应性炎症的作用。
Objective To investigate the effect of KeChuanLuo, a traditional Chinese medicine, on soluble intercellular adhesion molecule-1 (sICAM-1), eosinophils and eosinophil cationic protein (ECP) in asthmatic guinea pigs, and to elucidate the effect of cough and asthma treatment. Asthma anti-airway allergic inflammation mechanism of action. Methods The levels of ECP, sICAM-1 in plasma and eosinophils counts in bronchoalveolar lavage fluid of asthmatic guinea pigs were determined by fluorescence enzyme labeling, double antibody sandwich ELISA and density gradient cell separation. Results The contents of ECP and sICAM-1 in asthmatic guinea pigs and the total number of cells and low density eosinophils in alveolar lavage fluid were significantly higher than those in the normal group (P < 0.01). After treatment with Mesoconazole, it was significantly decreased (P < 0.01) and was close to normal (P > 0.05). Conclusion Kechuanchuan drop regulates the level of interleukin-5, inhibits the recruitment of eosinophils in the airway and release of toxic granule proteins, thus exerting its anti-allergic effects on airway inflammation.
