目的观察姜黄素对高脂饮食大鼠总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)含量和caveolin-1表达的影响。方法将40只健康成年雄性大鼠分为正常对照组(普通饲料喂养)、高脂血症模型组(高脂饲料喂养)、姜黄素低剂量组(高脂饲料+姜黄素100mg/kg.bw/d)、姜黄素高剂量组(高脂饲料+姜黄素200mg/kg.bw/d)。分别于实验开始前1d、实验第8周末禁食12h后,测血清TC、TG、LDL-C含量的变化。同时Western-Blot印迹法检测大鼠血管壁caveolin-1表达的变化。结果与正常对照组比较,高脂血症模型组的TC、TG、LDL-C分别由对照组的(1.58±0.25)、(0.55±0.06)、(0.49±0.06)mmol/L升高到高脂饮食组的(3.47±0.35)、(0.92±0.07)、(0.88±0.04)mmol/L,并伴随caveolin-1蛋白表达水平下降3.25倍。姜黄素的介入能降低高脂饮食大鼠TC、LDL-C、TG的含量以及促进其caveolin-1的表达(P<0.05)。结论姜黄素具有降低高脂饮食大鼠TC、LDL-C、TG含量的作用,其作用机制可能与促进caveolin-1表达有关。
Objective To observe the effect of curcumin on the content of total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C) and the expression of caveolin-1 in high fat diet rats. Methods Forty healthy adult male rats were divided into normal control group (fed with normal diet), hyperlipidemia model group (fed with high fat diet), curcumin low dose group (high fat diet + curcumin 100 mg / kg.bw / d), curcumin high-dose group (high fat diet + curcumin 200mg / kg.bw / d). The levels of TC, TG and LDL-C in serum were measured at 1 day before the experiment and at the end of the 8th week after fasting for 12 hours respectively. At the same time, the expression of caveolin-1 in rat vascular wall was detected by Western blotting. Results Compared with the normal control group, the levels of TC, TG and LDL-C in hyperlipidemia model group were increased to (1.58 ± 0.25) and (0.55 ± 0.06) and (0.49 ± 0.06) mmol / L (3.47 ± 0.35), (0.92 ± 0.07), (0.88 ± 0.04) mmol / L, respectively, with the decrease of caveolin-1 protein expression 3.25-fold. The intervention of curcumin reduced the content of TC, LDL-C, TG and the expression of caveolin-1 in high fat diet rats (P <0.05). Conclusion Curcumin can reduce the contents of TC, LDL-C and TG in rats fed high-fat diet. Its mechanism may be related to the promotion of caveolin-1 expression.
