目的:研究MMP-9及TIMP-3 mRNA在实验性小鼠CNV形成过程中的表达,探讨二者在CNV发生机制中的作用。方法:使用半导体二极管激光器(810nm)对C57BL/6J小鼠视网膜进行照射(120mW,75μm,0.1s)以诱导CNV形成,分别于激光后1,3d;1,2,4wk取出眼球,采用原位杂交技术及图象分析对MM-9、TIMP-3 mRNA的表达情况进行检测。结果:MMP-9、TIMP-3 mRNA表达在CNV形成过程中均有一动态过程,MMP-9 mRNA水平1,2,4wk>3d>1d(P<0.05),而TIMP-3mRNA表达3d;1,2,4wk>1d(P<0.05)。结论:MMP-9与TIMP-3 mRNA表达变化的失衡促使基质降解作用的发挥,在CNV的形成中可能发挥了一定作用。
OBJECTIVE: To study the expression of MMP-9 and TIMP-3 mRNA in the process of CNV formation in experimental mice and to explore their roles in the pathogenesis of CNV. Methods: The retina of C57BL / 6J mice was irradiated with a diode laser (810nm) to induce CNV formation. The eyeballs were removed at 1, 3, Hybridization and image analysis of MM-9, TIMP-3 mRNA expression were detected. Results: The mRNA expression of MMP-9 and TIMP-3 both had a dynamic process during the formation of CNV. The levels of MMP-9 mRNA were 1, 2 and 4wk> 3d> 1d (P <0.05) 2,4wk> 1d (P <0.05). Conclusion: The imbalance of MMP-9 and TIMP-3 mRNA expression promotes the degradation of stroma and may play a role in the formation of CNV.
