目的评价锰超氧化物歧化酶(MnSOD)基因Val-9Ala多态性与乳腺癌易感性及经期状态的关系。方法计算机检索Cochrane Library(2010年第1期)、PubMed、CNKI、CBM和万方数据库,按照纳入与排除标准选择相关的病例对照试验,检索时限均为建库至2010年10月。在提取资料和评价质量后,采用RevMan 4.2.10软件进行Meta分析。结果共纳入14个研究,合计17842例患者。Meta分析结果显示,MnSOD基因Val-9Ala多态性与乳腺癌易感性无关[Val/Ala vs.Val/Val:OR=1.04,95%C(I0.93,1.17);Ala/Ala vs.Val/Val:OR=1.12,95%C(I0.95,1.33);Ala/Ala vs.Val/Ala+Val/Val:OR=1.06,95%CI(0.93,1.20);Val/Ala+Ala/Ala vs.Val/Val:OR=1.06,95%CI(0.94,1.10)]。进一步亚组分析结果显示,MnSOD Val/Ala及Ala/Ala基因型会增加绝经前妇女患乳腺癌的风险[Val/Ala+Ala/Ala vs.Val/Val:OR=1.15,95%CI(1.01,1.31)]。结论 MnSOD基因Val-9Ala多态性与乳腺癌易感性无关,但可能增加绝经前妇女患乳腺癌的风险。
Objective To evaluate the relationship between Val-9Ala polymorphism of manganese superoxide dismutase (MnSOD) gene and breast cancer susceptibility and menstrual status. Methods The Cochrane Library (2010 issue 1), PubMed, CNKI, CBM and Wanfang database were searched by computer. According to the included case-control trial of inclusion and exclusion criteria, the search period was from the database to October 2010. After extracting data and evaluating the quality, RevMan 4.2.10 software was used for meta-analysis. Results A total of 14 studies were included, with a total of 17,842 patients. Meta analysis showed that Val-9Ala polymorphism of MnSOD gene was not associated with susceptibility to breast cancer [Val / Ala vs. Val / Val: OR = 1.04,95% C (I0.93,1.17); Ala / Ala vs. Val /Val:OR=1.12,95%C(I0.95,1.33);Ala/Ala vs.Val / Ala + Val / Val: OR = 1.06,95% CI (0.93,1.20); Val / Ala + Ala / Ala vs. Val / Val: OR = 1.06, 95% CI (0.94, 1.10)]. Further subgroup analyzes showed that MnSOD Val / Ala and Ala / Ala genotypes increased the risk of breast cancer in premenopausal women [Val / Ala + Ala vs. Ala / Val / Val: OR = 1.15, 95% CI , 1.31)]. Conclusion Val-9Ala polymorphism of MnSOD gene has nothing to do with breast cancer susceptibility, but may increase the risk of breast cancer in premenopausal women.
