目的 利用新生鼠缺氧缺血性脑损伤动物模型,探讨缺血缺氧后不同时间热休克蛋白(HSP70)的表达。方法采用阻断左侧颈总动脉制作新生鼠缺氧缺血性脑损伤动物模型,用原位杂交的技术观察正常对照组及缺氧缺血1 h、24 h、48 h、96h组HSP70表达变化以及阳性染色细胞的分布部位及数量。结果核酸原位杂交DAB染色、图象扫描分析显示,HSP70表达在缺氧缺血后1 h后即有升高,24 h时阳性表达最高,48 h、96 h表达逐渐降低。结论 脑缺氧缺血后热休克蛋白(HSP)表达增加,与脑缺氧缺血后神经细胞保护作用有关。
Objective To investigate the expression of heat shock protein (HSP70) at different time points after hypoxia-ischemia in neonatal rats with hypoxic-ischemic brain damage. Methods Animal models of neonatal hypoxic-ischemic brain damage were made by blocking the common carotid artery. The expression of HSP70 in normal control group, hypoxia-ischemia group 1 h, 24 h, 48 h, 96 h was observed by in situ hybridization Changes and positive staining cells and the distribution of the number of sites. Results Nucleic acid in situ hybridization (DAB) staining and image scanning analysis showed that the expression of HSP70 increased at 1 h after hypoxia-ischemia and reached the peak at 24 h, and gradually decreased at 48 h and 96 h. Conclusion The expression of heat shock protein (HSP) increased after hypoxic-ischemic brain injury, which is related to the neuroprotective effect after hypoxic-ischemic brain damage.
