目的:观察氧化低密度脂蛋白(oxldl)及其受体血凝素样氧化低密度脂蛋白受体1(lox-1)在运动影响动脉粥样硬化形成中的变化。方法:对8周龄ApoE基因敲除(ApoE-/-)小鼠饲以“西方饮食”饲料建立动脉粥样硬化模型,随机分为ApoE-/-安静组和ApoE-/-运动组。C57BL/6J小鼠为同源对照鼠,分为正常安静组和正常运动组。每组15只。运动小鼠进行12周中等强度跑台运动(10米/分×30分钟/天~13米/分×60分钟/天,每周5天)。采用ELISA及免疫组化方法检测血oxldl、lox-1的表达。结果:和正常安静组相比,ApoE-/-小鼠oxldl显著升高(P<0.01);ApoE-/-运动组oxldl较ApoE-/-安静组降低(P<0.05)。正常安静组和正常运动组血oxldl无显著性差异。正常安静组主动脉lox-1弱表达,ApoE-/-安静组病变内皮层、斑块中可见明显的棕黄色颗粒、棕黄色弧线,ApoE-/-运动组表达较ApoE-/-安静组明显减弱。经图像分析,正常安静组、正常运动组主动脉弓lox-1阳性表达面积和阳性面积百分比组间比较均无显著差异。和正常安静组相比,ApoE-/-安静组阳性表达面积显著增大(P<0.01)。ApoE-/-运动组阳性表达面积显著低于ApoE-/-安静组(P<0.01)。结论:12周有氧运动可下调ApoE-/-小鼠血oxldl水平,显著降低其受体lox-1表达。
AIM: To observe the changes of oxidative low density lipoprotein (oxLDl) and its receptor, hemagglutinin-like oxidized low density lipoprotein receptor 1 (lox-1), in the process of exercise-induced atherosclerosis. Methods: ApoE - / - mice of 8 weeks old were fed with “Western diet” to establish atherosclerosis model and were randomly divided into ApoE - / - Quiet group and ApoE - / - exercise group . C57BL / 6J mice were homologous control mice, divided into normal quiet group and normal exercise group. 15 in each group Exercise mice were subjected to 12 weeks of moderate treadmill exercise (10 m / min x 30 min / day ~ 13 m / min x 60 min / day, 5 days per week). The expression of oxLDL and lox-1 in blood was detected by ELISA and immunohistochemistry. Results: Compared with normal quiet group, oxLDL of ApoE - / - mice was significantly increased (P <0.01); oxLDL of ApoE - / - exercise group was lower than that of ApoE - / - quiet group (P <0.05). There was no significant difference in oxLDL between normal quiet group and normal exercise group. In the normal and quiet group, lox-1 was weakly expressed in the apoE - / - quiescent group, and obvious brown granules were seen in the plaques of the ApoE - / - quiescent group. Compared with the ApoE - / - quiescent group Obviously weakened. The image analysis, normal quiet group, normal exercise group aortic arch lox-1 positive expression area and positive area percentage comparison between the two groups showed no significant difference. Compared with the normal quiet group, the positive expression area of ApoE - / - Quiet group was significantly increased (P <0.01). The positive expression area of ApoE - / - exercise group was significantly lower than that of ApoE - / - Quiet group (P <0.01). CONCLUSION: Aerobic exercise at 12 weeks reduces the level of oxLDL in ApoE - / - mice and significantly decreases the expression of its receptor.
