随着陆地资源的减少,海洋资源为药物开发提供了一条新途径,其中海绵是重要的海洋生物资源之一。近年来,人类已从海绵中分离提取出许多结构新颖的抗肿瘤活性成分,海绵靶向抗肿瘤药物的开发成为开发海洋资源创新药物的新趋势。该文根据海绵抗肿瘤活性成分及其相关合成类似物的作用靶点,包括微管与微丝等细胞骨架系统、细胞周期素蛋白与极光激酶等蛋白激酶家族、DNA合成及其相关酶、肿瘤组织生长微环境、人体免疫系统等,对海绵活性成分及其相关合成物的抗肿瘤作用机制进行综述,并简单介绍其相关临床研究情况。
With the reduction of land resources, marine resources provide a new way for drug development. Among them, sponges are one of the important marine living resources. In recent years, mankind has isolated and extracted many novel anti-tumor active ingredients from sponges. The development of sponge targeted anti-tumor drugs has become a new trend of developing innovative drugs for marine resources. According to the target of sponge antitumor activity and its related synthetic analogs, this article includes cytoskeleton system such as microtubules and microfilaments, protein kinase family such as cyclin and aurora kinase, DNA synthesis and related enzymes, Tissue growth microenvironment, the human immune system, the sponge active ingredients and their related compounds antitumor mechanisms are reviewed, and briefly introduce the relevant clinical research.
