目的:建立同时检测不同品种、不同产地蜈蚣样品中8种核苷类成分(胞嘧啶,胞苷,次黄嘌呤,黄嘌呤,尿苷,胸腺嘧啶,2’-脱氧肌苷和胸苷)含量的高效液相色谱法。方法:不同品种及产地蜈蚣提取物的分析选用Hedera ODS-2 C_(18)(4.6 mm×250 mm,5μm)色谱柱,流动相为甲醇-水,梯度洗脱,流速1.0 m L·min~(-1),柱温30℃,检测波长254 nm,进样量5μL。结果:8种核苷类成分分离良好;在一定质量浓度内线性关系良好(r>0.999 9);加样回收率在95.73%~102.6%;11个批次蜈蚣样品中均含有8种核苷类成分,但含量存在差异。不同品种、不同产地的蜈蚣中核苷成分总量墨江蜈蚣含量最高,多棘蜈蚣其次,少棘巨蜈蚣和黑头蜈蚣中核苷类总成分比较接近。蜈蚣中次黄嘌呤和黄嘌呤明显高于其他核苷成分,少棘巨蜈蚣中次黄嘌呤的含量要高于其他品种。结论:该文所建立的方法快速简便,重复性、准确度良好,为蜈蚣的质量控制提供依据。
OBJECTIVE: To establish a method for the simultaneous determination of 8 nucleosides (cytosine, cytidine, hypoxanthine, xanthine, uridine, thymine, 2’-deoxyinosine and thymidine) in centipede samples of different varieties and origin High Performance Liquid Chromatography. Methods: Hedera ODS-2 C_ (18) (4.6 mm × 250 mm, 5 μm) column was used for the analysis of centipede extracts from different varieties and producing areas. The mobile phase consisted of methanol-water with gradient elution at a flow rate of 1.0 m L · min ~ (-1), column temperature 30 ℃, detection wavelength 254 nm, injection volume 5μL. Results: The eight nucleosides were well separated. The linearity was good (r> 0.999 9) within a certain mass concentration. The average recoveries were 95.73% ~ 102.6%. The 11 batches of centipede samples contained 8 nucleosides Class components, but there are differences in content. Centipede in different varieties, different origin of the total amount of the nucleoside content Mojiang highest centipede, spiny centipede secondly, spines giant centipede black head centipede nucleosides in the total composition is relatively close. In centipede, hypoxanthine and xanthine were significantly higher than those of other nucleosides, and the content of hypoxanthine in spiky centipede was higher than that of other species. Conclusion: The method established in this paper is rapid, simple, reproducible and accurate, providing the basis for the quality control of centipede.
