目的检查模拟失重对大鼠比目鱼肌梭内肌纤维肌球蛋白重链表达的影响 ,探讨模拟失重所致肌梭退变的机制。方法采用免疫过氧化酶反应ABC法观察尾悬吊 7d ,1 4d与对照组大鼠比目鱼肌梭内肌纤维对特异性抗小鸡肌球蛋白重链慢紧张型 (ALD5 8)和快缩型 (MF30 )单克隆抗体表达的差别。结果梭外肌纤维不与ALD5 8和MF30起反应。这两种抗体仅与梭内肌纤维反应。核袋 1和核袋 2纤维对ALD5 8抗体起反应。核袋 2和核链纤维对MF30抗体起反应。尾悬吊 7d后 ,核袋 1和核袋 2纤维对ALD5 8的免疫反应性降低 ,而核袋 2和核链纤维对MF30免疫反应性增强。尾悬吊 1 4d后 ,核袋 1和核袋 2纤维对ALD5 8的免疫反应性明显降低 ,甚至消失。而所有的梭内肌纤维对MF30都起免疫反应 ,并且明显增强。结论模拟失重可致大鼠比目鱼肌梭内肌纤维肌球蛋白重链的表达水平和表型发生改变
Objective To investigate the effect of simulated weightlessness on the expression of myosin heavy chain in the myofibrillar muscle of the soleus muscle of rats and to explore the mechanism of simulated muscle spindle degeneration induced by weightlessness. Methods Immuno-peroxidase (ABC) method was used to observe the effects of the myofusoid fibronectin on the specific anti-chicken myosin heavy chain slow strain (ALD5 8) and rapid constriction (7d, 14d) MF30) monoclonal antibody expression differences. Results Extrasolar muscle fibers did not react with ALD5 8 and MF30. Both antibodies react only with the intrafusal myofibers. Nucleus 1 and 2 were reactive with ALD5 8 antibody. Nuclear bag 2 and nuclear chain fibers react to the MF30 antibody. After the tail suspension for 7 days, the immunoreactivity of nuclear bag 1 and nuclear bag 2 fibers to ALD5 8 decreased, whereas the nuclear bag 2 and nuclear chain fibers increased the immunoreactivity to MF30. After the tail suspension for 14 days, the immunoreactivity of nuclear bag 1 and nuclear bag 2 fibers to ALD5 8 was significantly reduced or even disappeared. All of the intrafusal myofibers were immunoreactive with MF30 and markedly enhanced. Conclusion Simulated weightlessness can change the expression and phenotype of myosin heavy chain in the soleus muscle of the soleus muscle in rats
