目的从兔骨骼肌中提取肌球蛋白轻链,为下一步研究肌肉收缩原理提供可磷酸化的底物。方法用离心、透析、柱层析、SDS聚丙烯酰胺凝胶电泳(SDS-PAGE)和电泳转移等方法纯化兔骨骼肌肌球蛋白轻链。结果提取纯化得到较纯的肌球蛋白,进一步得到轻链混合液和各种轻链纯品,经紫外吸收检测及SDS0PAG证实其分子量分别为25000(MLC1)、19000(MLC2)和16500(MLC3),均与文献报道基本一致。结论研究所用的方法可行,所得到的肌球蛋白及其轻链样品较纯,可用于下一步实验。
Objective To extract myosin light chain from rabbit skeletal muscle and provide phosphorylated substrate for further study of muscle contraction. Methods Rabbit skeletal muscle myosin light chain was purified by centrifugation, dialysis, column chromatography, SDS polyacrylamide gel electrophoresis (SDS-PAGE) and electrophoretic transfer. Results The pure myosin was extracted and purified, and the light chain mixture and various light chain purified products were further obtained. The molecular weights of MLC1, MLC2 and MLC3 were 25000 (MLC1), 25000 (MLC1) , Are basically the same as reported in the literature. Conclusion The method used in this study is feasible. The resulting myosin and light chain samples are purer and can be used in the next experiment.
