The authors outline the history of sporozoites producing protective immunity and highlight the role of circumspo-rozoite proteins (CSPs) in the development of sporozoite vaccines. The outer membrane of all mature and infectious salivary gland sporozoites is covered by an acidic 30-60 kDa polypeptide called the circumsporozoite protein (CSP). CSP monoclonal antibodies and their antibody-binding fragments (Fabs) neutralize the infectivity of sporozoites in vivo and in vitro and prevent sporozoite attachment and entry into target cells.
