目的 :观察大鼠脑出血后神经缺损症状和线粒体膜电位的改变 ,及中药制剂平肝熄风汤的干预作用。方法 :用Ⅶ型胶原酶于脑内定位注射诱导大鼠脑出血模型。采用神经缺损积分评定神经功能 ;用荧光分光光度计测定线粒体悬液的荧光强度反映线粒体膜电位。结果 :大鼠脑出血后迅速出现神经缺损症状 ,平肝熄风汤能明显降低神经缺损症状积分。脑出血大鼠线粒体膜电位于术后 4h显著下降 ,1d时膜电位下降达高峰 ,3,7d逐步恢复 ,仍低于正常组和假手术组 (P <0 .0 5 )。平肝熄风汤治疗组在脑出血后 1d ,3d ,7d ,线粒体膜电位均明显高于模型组 (P <0 .0 5 )。结论 :大鼠脑出血后线粒体膜电位下降 ;平肝熄风汤能阻断膜电位下降 ,同时能减轻出血后神经元损伤。
Objective : To observe the neurological deficits and changes of mitochondrial membrane potential after intracerebral hemorrhage in rats, and the intervention of Pinggan Xifeng Decoction, a traditional Chinese medicine preparation. Methods : A rat model of intracerebral hemorrhage induced by local injection of type VII collagenase was established. Neurological deficits were used to assess neural function; fluorescence intensity of mitochondrial suspensions was measured with a fluorescence spectrophotometer to reflect mitochondrial membrane potential. Results: The symptoms of neurological deficits occurred rapidly after cerebral hemorrhage in rats. Pinggan Xifeng Decoction can significantly reduce the symptoms of neurological deficits. The mitochondrial membranes of rats with intracerebral hemorrhage were significantly decreased at 4 h postoperatively. The membrane potential decreased to a peak at 1d and gradually recovered at 3 and 7d, which was still lower than that in the normal and sham groups (P < 0.05). In the Pinggan Xifeng Decoction group, the mitochondrial membrane potential was significantly higher than that of the model group 1 d, 3 d, and 7 d after cerebral hemorrhage (P < 0.05). Conclusion: The mitochondrial membrane potential decreased after intracerebral hemorrhage in rats. Pinggan Xifeng Decoction can block the decrease of membrane potential and reduce neuronal damage after hemorrhage.
