目的:探讨醋酸棉酚(gossypol acetic acid,GAA)对人舌鳞癌Tca8113细胞体外生长、凋亡以及DNA甲基转移酶1(DNA methyltransferase 1,DNMT1)mRNA表达的影响。方法:不同浓度GAA作用Tca8113细胞后,MTT法和FCM法检测细胞增殖和细胞凋亡,实时荧光定量PCR法检测Tca8113细胞中DNMT1mRNA的表达水平。结果:GAA作用Tca8113细胞24、48和72h后,细胞的生长受到抑制;30μmol/LGAA作用48h和15μmol/LGAA作用72h后,Tca8113细胞的凋亡率明显高于GAA未作用的对照组(P<0.05);5、10、15和20μmol/LGAA作用Tca8113细胞48h后,细胞中DNMT1mRNA的表达水平低于对照组(P<0.05)。结论:GAA能抑制人舌鳞癌Tca8113细胞的生长、诱导细胞凋亡,并下调Tca8113细胞中DNMT1mRNA的表达水平。
AIM: To investigate the effects of gossypol acetic acid (GAA) on the growth and apoptosis of human tongue squamous cell carcinoma Tca8113 cells and the expression of DNA methyltransferase 1 (DNMT1) mRNA. Methods: Tca8113 cells were treated with different concentrations of GAA. Cell proliferation and apoptosis were detected by MTT and FCM. The expression of DNMT1 mRNA in Tca8113 cells was detected by real-time fluorescence quantitative PCR. Results: The growth of Tca8113 cells was inhibited by GAA for 24, 48 and 72 hours. The apoptotic rates of Tca8113 cells treated with 30μmol / L GAA for 48h and 15μmol / L GAA were significantly higher than those without GAA (P < 0.05). After treated with 5,10,15 and 20μmol / L GAA for 48h, the expression of DNMT1 mRNA in Tca8113 cells was lower than that in control group (P <0.05). CONCLUSION: GAA can inhibit the growth of human tongue squamous cell carcinoma Tca8113 cells, induce apoptosis and down-regulate the expression of DNMT1 mRNA in Tca8113 cells.
