目的探讨高浓度葡萄糖对人肾脏近曲小管上皮(HK-2)细胞的糖毒性及α-亚麻酸/亚油酸(ALA/LA)的改善作用。方法常规方法培养接种HK-2细胞,设置11个葡萄糖浓度,MTT法检测葡萄糖的糖毒性;设立正常对照组、高糖模型组、阳性对照组、ALA组、LA组和ALA/LA组,检测a-ALA/LA的干预作用;葡萄糖氧化酶-过氧化物酶(GOD-POD)法检测HK-2细胞的葡萄糖吸收。结果高浓度葡萄糖(≥25.3 mmol/L,培养48h)显著抑制HK-2细胞的存活率(P<0.05);培养48 h,葡萄糖浓度25.3~56.5 mmol/L可作为HK-2细胞葡萄糖毒性造模条件;适宜剂量的ALA、LA及适宜比例的ALA/LA,能够降低HK-2细胞的葡萄糖毒性,增加HK-2细胞的葡萄糖吸收。结论必需脂肪酸ALA/LA能降低葡萄糖对HK-2细胞的糖毒性,增加其葡萄糖吸收功能。
Objective To investigate the effects of high glucose on the glucose tolerance of human proximal tubular epithelial cells (HK-2) and the effects of α-linolenic acid and linoleic acid (ALA / LA). Methods HK-2 cells were inoculated with HK-2 cells in a routine manner and glucose concentrations were determined by MTT assay. The normal control group, high glucose model group, positive control group, ALA group, LA group and ALA / a-ALA / LA intervention. Glucose oxidase-peroxidase (GOD-POD) was used to detect the glucose uptake in HK-2 cells. Results Glucose (≥25.3 mmol / L, cultured for 48 h) significantly inhibited the viability of HK-2 cells (P <0.05). After cultured for 48 h, glucose concentrations of 25.3-56.5 mmol / Modal conditions; the appropriate dose of ALA, LA and a suitable proportion of ALA / LA, can reduce the glucose toxicity of HK-2 cells and increase the glucose absorption of HK-2 cells. Conclusion ALA / LA, an essential fatty acid, can reduce the glucose toxicity of HK-2 cells and increase its glucose uptake.
