以籼稻‘双桂1号’(热敏感品种)和‘黄华占’(耐热品种)为材料,研究减数分裂期高温胁迫对水稻幼穗内源激素、多胺积累和蛋白质表达的影响。结果表明:在高温下,‘黄华占’的受精率和饱粒率显著高于‘双桂1号’。高温引起了两品种幼穗中吲哚乙酸(IAA)及玉米素(Z)和玉米素核苷(ZR)减少,脱落酸(ABA)含量增加,且‘黄华占’的变化小于‘双桂1号’;高温使两品种精胺(Spm)含量增加,‘双桂1号’的腐胺(Put)和亚精胺(Spd)含量减少,但‘黄华占’Put和Spd含量增加。两品种蛋白质表达对高温响应有差异,这些差异蛋白质主要涉及氧化还原调控、糖酵解、氨基酸代谢、光系统和胁迫响应。‘黄华占’的氧化还原调控蛋白在高温下表达量较高。表明耐热品种在高温下受精结实较好与其较高的IAA、Z+ZR和游离多胺含量,较低的ABA含量及高表达的氧化还原调控相关的蛋白有密切关系。
Effects of high temperature stress on the endogenous hormones, polyamine accumulation and protein expression in young panicles of rice at indica rice ’Shuanggui No.1’ (heat-sensitive variety) and ’Huanghuazhan’ (heat-tolerant variety) were studied. The results showed that under high temperature, the fertilization rate and the rate of grain filling of ’Huanghuazhan’ were significantly higher than that of ’Shuanggui No.1’. High temperature caused the decrease of IAA, Z and ZR and the increase of abscisic acid (ABA), and the change of ’Huanghuazhan’ was less than that of ’Shuanggui No.1’ ’. High temperature increased the content of spermine (Spm) in two cultivars and decreased Put and Spd contents in Shuiui 1, but increased Put and Spd in Huanghuazhan. The differences of protein expression between the two cultivars in response to high temperature were found. These differential proteins mainly involved in redox regulation, glycolysis, amino acid metabolism, photosystem and stress response. ’Huang Hua Zhan’ redox regulatory protein expression at high temperatures higher. The results showed that fertilization and seed setting of heat-tolerant cultivars at high temperature was closely related to their higher contents of IAA, Z + ZR and free polyamines, lower contents of ABA and higher expression of redox-regulated proteins.
