目的研究太白银莲花皂甙-1对人脑胶质母细胞瘤U87MG凋亡的影响,并探讨其可能的分子机制。方法采用四甲基偶氮唑蓝(MTT)比色法检测不同浓度不同时间(6、24、48、72 h)太白银莲花皂甙-1对U87MG细胞生长增殖的抑制作用;应用倒置显微镜和Hoehst荧光染色、末端脱氧核苷酸转移酶介导的生物素脱氧尿嘧啶核苷酸缺口末端标记法(TUNEL)、膜联蛋白V-异硫氰酸荧光素/碘化丙啶(AnnexinV-FITC/PI)检测太白银莲花皂甙-1作用U87MG细胞后形态的变化和凋亡情况;通过免疫印迹法法检测太白银莲花皂甙-1处理后细胞凋亡相关蛋白Survivin及活化的Caspase-3的表达。结果太白银莲花皂甙-1可明显抑制U87MG细胞的增殖,诱导其凋亡;同时下调Survivin表达,激活了凋亡蛋白Activated Caspase-3的表达。结论体外实验显示,太白银莲花皂甙-1对U87MG细胞具有显著的增殖抑制作用,并能成功诱导人脑胶质母细胞瘤U87MG细胞发生凋亡,其机制可能与抑制细胞增殖及诱导细胞凋亡有关。
Objective To investigate the effect of Aloe saginata-1 on the apoptosis of human glioblastoma U87MG cells and to explore its possible molecular mechanism. Methods MTT assay was used to detect the growth inhibition of U87MG cells at different concentrations (6,24,48 and 72 h) of Taibaihua lotus saponin-1. The growth of U87MG cells was observed by inverted microscope and Hoehst Fluorescence staining, terminal deoxynucleotidyl transferase-mediated biotin deoxyuridine nucleotide nick end labeling (TUNEL), annexin V-FITC / PI) were used to detect the morphological changes and apoptosis of U87MG cells. The expressions of Survivin and Caspase-3 were detected by Western blotting. Results Taibai lotus saponin-1 significantly inhibited the proliferation of U87MG cells and induced its apoptosis. At the same time, the expression of Survivin was down-regulated and the expression of Activated Caspase-3 was activated. Conclusions In vitro experiments showed that Taibai lotus saponin-1 could significantly inhibit the proliferation of U87MG cells and induce the apoptosis of U87MG cells, which may be related to the inhibition of cell proliferation and induction of apoptosis related.
