将人胰岛素样生长因子-1(IGF-1)重组腺病毒(Ad IGF-1)转染C17.2神经干细胞;Western blot法检测IGF-1蛋白的表达;建立神经干细胞缺氧模型,TUNEL法检测细胞凋亡指数,流式细胞术测定细胞凋亡率的变化。发现转染Ad IGF-1的C17.2神经干细胞成功表达IGF-1蛋白;缺氧诱导C17.2神经干细胞凋亡,转染Ad IGF-1的C17.2神经干细胞凋亡指数、凋亡率与对照组相比有显著差异。提示转染Ad IGF-1能减轻缺氧诱导的C17.2神经干细胞凋亡。
C17.2 neural stem cells were transfected with human IGF-1 recombinant adenovirus (Ad-IGF-1), the expression of IGF-1 protein was detected by Western blot, and the neural stem cell hypoxia model was established by TUNEL method The apoptosis index was detected and the change of apoptosis rate was determined by flow cytometry. The results showed that IGF-1 protein was successfully expressed in C17.2 neural stem cells transfected with Ad IGF-1; apoptosis of C17.2 neural stem cells induced by hypoxia; apoptotic index of C17.2 neural stem cells transfected with Ad IGF-1; Compared with the control group, there are significant differences. These results suggest that transfection of Ad-IGF-1 can reduce hypoxia-induced apoptosis of C17.2 neural stem cells.
