目的 :研究正常生理状态下钙调蛋白 (calmodulin)的结构特征和分布规律。方法 :本研究应用免疫细胞化学方法 ,采用单克隆抗体及免疫荧光标记技术 ,借助于荧光显微镜和激光扫描共聚焦显微镜 ,逐层观察耳蜗钙调蛋白的分布特点。结果 :豚鼠耳蜗外毛细胞 (OHCs)和内毛细胞 (IHCs)各转之间 calmodulin分布无变化 ,OHCs内 calmodulin含量多于 IHCs。OHCs表皮板和静纤毛出现 calmodulin特异性着色 ,表明哺乳动物耳蜗 OHCs的静纤毛含有 calmodulin。支持细胞内无 calmodulin的分布。结论 :豚鼠耳蜗 OHCs、IHCs和支持细胞中 calmodulin的结构和分布有显著差异。
Objective: To study the structural characteristics and distribution of calmodulin under normal physiological conditions. Methods: Immunocytochemistry was used in this study. The distribution characteristics of cochlear calmodulin were observed by fluorescence microscopy and laser scanning confocal microscopy with monoclonal antibody and immunofluorescence labeling. Results: There was no change in calmodulin distribution between guinea pig cochlear outer hair cells (OHCs) and inner hair cells (IHCs), and the content of calmodulin in OHCs was higher than that in IHCs. Calmodulin-specific staining of OHCs epidermal and ciliary fibers revealed calmodulin expression in the calluses of mammalian cochlear OHCs. Supports intracellular calmodulin distribution. Conclusion: The structure and distribution of calmodulin in OHCs, IHCs and supporting cells of guinea pig cochlea are significantly different.
