目的:研究GnRHα控制性超促排卵对围着床期小鼠子宫内膜溶血磷脂酸受体3(LPAR3)mRNA和蛋白表达的影响。方法:采用RT-PCR、Western blot方法检测自然受孕组(NC)和超促排卵组(COH)小鼠性交后第3~6天(3~6d.p.c)子宫内膜LPAR3mRNA和蛋白表达的表达。应用免疫化学发光法测定血清雌、孕激素浓度。结果:COH组3d.p.cLPAR3mRNA和蛋白的表达最强,NC组4d.p.c表达最强。其余相应各时间点两组间表达量差异无统计学意义;雌、孕激素在两组均随着妊娠天数增加而升高。相应同期各时间点COH组高于NC组(P<0.05)。结论:COH可能通过改变与子宫内膜容受性相关的LPAR3的时序变化影响子宫内膜容受性,从而影响胚胎着床。
Objective: To investigate the effect of GnRHα controlled ovarian hyperstimulation on mRNA and protein expression of LPAR3 in peri-implantation mouse. Methods: The expression of LPAR3 mRNA and protein in endometrium of 3 and 6 days (3 ~ 6 days) postnatal mice in NC group and COH group were detected by RT-PCR and Western blot respectively . Serum estrogen and progesterone concentrations were determined by immunochemiluminescence. Results: The expression of 3d.p.cLPAR3mRNA and protein in COH group was the strongest, and the expression of 4d.p.c was the strongest in NC group. The rest of the corresponding time points there was no significant difference between the two groups expression; estrogen and progesterone in both groups increased with the number of gestational days increased. The corresponding COH group was higher than NC group at each time point (P <0.05). CONCLUSIONS: COH may affect endometrial receptivity by changing the temporal change of LPAR3 associated with endometrial receptivity, which may affect embryo implantation.
