目的探讨丙戊酸钠(VPA)对多发性骨髓瘤(MM)细胞株细胞凋亡与自噬的影响。方法 MM细胞株RPMI8226和U266分为空白对照组及VPA 2、4、8mmol/L三个处理组。HE染色法观察VPA诱导细胞凋亡的形态学改变;流式细胞术检测VPA诱导RPMI8226和U266细胞凋亡情况。实时定量PCR和Western blot检测凋亡信号传导通路蛋白Bcl-2、Bax、Caspase-3和Caspase-8 mRNA和蛋白表达。结果随着VPA浓度升高,RPMI8226和U266细胞凋亡率上升(P<0.05)。VPA浓度为8mmol/L时,Bax、Caspase-3、Caspase-8 mRNA和蛋白相对表达量随着VPA作用时间延长而增加,而Bcl-2 mRNA和蛋白相对表达量随VPA作用时间延长而下降(P<0.05)。结论 VPA促进MM细胞株RPMI8226和U266细胞凋亡,且呈浓度和时间依赖性。
Objective To investigate the effect of VPA on apoptosis and autophagy in multiple myeloma (MM) cell lines. Methods MM cell lines RPMI8226 and U266 were divided into blank control group and VPA 2,4,8mmol / L three treatment groups. The morphological changes of VPA-induced apoptosis were observed by HE staining. The apoptosis of RPMI-8226 and U266 cells was detected by flow cytometry. The mRNA and protein expressions of Bcl-2, Bax, Caspase-3 and Caspase-8 were detected by real-time PCR and Western blot. Results As the VPA concentration increased, the apoptosis rate of RPMI8226 and U266 cells increased (P <0.05). When the VPA concentration was 8mmol / L, the relative expression of Bax, Caspase-3 and Caspase-8 mRNA and protein increased with the prolongation of VPA, while the relative expression of Bcl-2 mRNA and protein decreased with the prolongation of VPA P <0.05). Conclusion VPA can promote the apoptosis of RPMI8226 and U266 cells in a concentration-and time-dependent manner.
