目的:研究高糖环境下人近端肾小管上皮细胞肌醇加氧酶(MIOX)的表达。方法:将人近端肾小管上皮细胞株HK-2分组培养,正常对照组培养基含5.5mmol/LD-葡萄糖,渗透浓度对照组培养基含19.5mmol/L甘露醇和5.5mmol/LD-葡萄糖,高糖组培养基含25mmol/LD-葡萄糖。分别于培养12、24、48、72h后取出爬片,应用免疫荧光细胞化学法检测MIOX蛋白的表达。结果:正常对照组和渗透浓度对照组HK-2细胞胞质MIOX蛋白微弱表达,各时间点表达量差异无统计学意义(P>0.05)。高糖组培养12h时,HK-2细胞MIOX蛋白表达即增高,随培养时间的延长,MIOX蛋白表达量逐渐升高,于48h达高峰。高糖组MIOX蛋白表达水平较正常对照组升高(P<0.01)。结论:高糖能促进人近端肾小管上皮细胞MIOX的表达,从而介导糖尿病患者体内肌醇耗竭。
AIM: To investigate the expression of myo-inositol oxygenase (MIOX) in human proximal tubular epithelial cells under high glucose conditions. Methods: Human proximal renal tubular epithelial cell line HK-2 was cultured in groups. The normal control group contained 5.5 mmol / L-glucose and the control group contained 19.5 mmol / L mannitol and 5.5 mmol / L-glucose. High glucose group culture medium containing 25mmol / LD-glucose. The slides were removed after culture for 12, 24, 48 and 72h, respectively. MIOX protein expression was detected by immunofluorescence cytochemistry. Results: The expression of MIOX in cytoplasm of HK-2 cells in normal control group and infiltrated control group was weakly expressed, with no significant difference at each time point (P> 0.05). The expression of MIOX protein in HK-2 cells increased at 12 h in high glucose group. The MIOX protein expression gradually increased with the increase of culture time and peaked at 48 h. MIOX protein expression in high glucose group was higher than that in normal control group (P <0.01). Conclusion: High glucose can promote the expression of MIOX in human proximal tubular epithelial cells, and thus mediate the inositol depletion in diabetic patients.
