目的 :研究不同载体对基因治疗效果的影响。方法 :采用本实验室建立的犬心肌缺血动物模型 ,通过冠状动脉造影和TTC染色两种方法比较了携带肝细胞生长因子基因的重组腺病毒Ad_HGF和质粒puDKH对心肌缺血的治疗效果 ;并且在体外采用绿色荧光蛋白报告基因测定了不同载体对原代心肌细胞的转染率。结果 :重组腺病毒Ad_HGF对心肌细胞的转染率和心肌缺血的治疗效果均好于重组质粒puDKH。结论 :导入系统是影响基因治疗效果的关键因素之一
Objective: To study the effect of different vectors on gene therapy. Methods: The animal models of canine myocardial ischemia established in our laboratory were used to compare the therapeutic effects of Ad_HGF and puDKH, a recombinant adenovirus carrying hepatocyte growth factor gene, on myocardial ischemia by coronary angiography and TTC staining. The green fluorescent protein reporter gene was used to determine the transfection efficiency of different vectors on primary cardiomyocytes in vitro. Results: The transfection efficiency of recombinant adenovirus Ad_HGF on cardiomyocytes and myocardial ischemia were better than that of the recombinant plasmid puDKH. Conclusion: The introduction of the system is one of the key factors affecting the effect of gene therapy
