目的 探讨全反式维甲酸 (ATRA)对人膀胱癌细胞T2 4的生长抑制和凋亡诱导作用。方法 应用细胞和分子生物学技术检测不同浓度ATRA对T2 4细胞生长和凋亡的影响。结果 3× 10 -5M~ 3× 10 -7MATRA可显著抑制T2 4细胞增殖。用 3× 10 -5M和 3× 10 -6MATRA作用细胞6天出现典型凋亡特征 ;3× 10 -5M、3× 10 -6M组及对照组的细胞凋亡率分别为 2 0 16%、15 31%和1 4 9% ;DNA琼脂糖凝胶电泳呈现凋亡特征性的DNA梯形带。结论 ATRA对人膀胱癌细胞T2 4有剂量生长抑制作用 ,且可成功地诱导人膀胱癌细胞T2 4发生凋亡。
Objective To investigate the effect of all-trans retinoic acid (ATRA) on the growth inhibition and apoptosis induction of human bladder cancer cell line T2 4. Methods Cell and molecular biology techniques were used to detect the effects of different concentrations of ATRA on the growth and apoptosis of T2 4 cells. Results 3 × 10 -5 M ~ 3 × 10 -7 MTATRA significantly inhibited the proliferation of T2 4 cells. The typical apoptotic characteristics of the cells treated with 3 × 10 -5 M and 3 × 10 -6 MATRA for 6 days were observed. The apoptosis rates of 3 × 10 -5 M, 3 × 10 -6 M and control groups were 20%, 15% 31% and 14.9%, respectively. The DNA ladder showed DNA ladder characteristic of DNA electrophoresis on DNA agarose gel electrophoresis. Conclusion ATRA can inhibit the growth of human bladder cancer cell line T2 4 in a dose-dependent manner and induce the apoptosis of human bladder cancer cell line T2 4 successfully.
