目的:建立中风通络丸中丹参酮ⅡA和丹酚酸B的含量同时测定法。方法:色谱柱为依利特Hypersil-ODS(4.6 mm×250 mm,5μm);流动相为乙腈-0.5%甲酸溶液;梯度洗脱:0~15 min,乙腈10%~20%;15~25 min,乙腈20%~50%;25~35 min,乙腈50%~60%;35~40 min,乙腈60%~95%,;流速为1.0 mL·min-1;检测波长:丹参酮ⅡA为270 nm,丹酚酸B为286 nm;柱温为30℃。结果:丹参酮ⅡA的线性方程为:Y=4×106X+175439(r=0.9998),线性范围为0.1861~0.4839μg,加样回收率为99.15%,RSD为1.20%;丹酚酸B的线性方程为:Y=2×106X+36551(r=0.9998),线性范围为0.6258~2.1902μg,加样回收率为100.19%,RSD为1.44%。结论:本方法操作简便,结果可靠,重现性好,可以用于中风通络丸的质量控制。
Objective: To establish simultaneous determination of tanshinone ⅡA and salvianolic acid B in Zhongfeng Tongluo Pills. METHODS: The column was Elite Hypersil-ODS (4.6 mm × 250 mm, 5 μm). The mobile phase consisted of acetonitrile-0.5% formic acid solution. The gradient elution was 0-15 min with 10-20% acetonitrile. , Acetonitrile 20% ~ 50%, 25 ~ 35 min, acetonitrile 50% ~ 60%, 35 ~ 40 min, acetonitrile 60% ~ 95%, the flow rate 1.0 mL · min-1; detection wavelength: tanshinone Ⅱ A 270 nm , Salvianolic acid B was 286 nm; column temperature was 30 ℃. Results: The linear equation of tanshinone ⅡA was: Y = 4 × 106X + 175439 (r = 0.9998) with a linear range of 0.1861 ~ 0.4839μg with the recovery of 99.15% and the RSD of 1.20% As follows: Y = 2 × 106X + 36551 (r = 0.9998), the linear range was 0.6258 ~ 2.1902μg, the recovery was 100.19% and the RSD was 1.44%. Conclusion: The method is simple, reliable, reproducible and can be used for the quality control of Zhongfeng Tongluo Pills.
