基于缩硫醛脱保护机制的比例计量型Hg~(2+)荧光探针及其斑马鱼活体造影应用

利用汞离子特异性诱导缩硫醛脱保护,引发分子内电荷转移发生改变的机制,设计合成了一种新型的激发型比例计量汞离子荧光探针.该探针在与汞离子结合后其最大激发波长由410 nm红移至485 nm,两个波长下的荧光强比值(F485/F410)由0.06增长至5.02.同时该探针对汞离子表现出了良好的比例计量响应选择性和较快的响应速度,并且检测限低于美国环境保护组织的饮用水标准.共聚焦造影研究发现,该探针可以应用于活细胞及5天龄斑马鱼幼体中的汞离子的检测.利用该性质对Hg2+在斑马鱼体内分布及毒性进行了初步的研究. A new type of excited proportional mercury ion fluorescence probe was designed and synthesized by using mercury ion to specifically induce the dethrination of thione and initiate the mechanism of charge transfer in the molecule. The excitation wavelength was red-shifted from 410 nm to 485 nm, and the fluorescence intensity ratio (F485 / F410) at both wavelengths increased from 0.06 to 5.02. At the same time, the probe showed a good selectivity and faster response to mercury ion , And the detection limit is lower than that of the US Environmental Protection Organization’s drinking water standard.Confocal contrast studies have found that the probe can be applied to the detection of mercury ions in living cells and 5-day-old zebrafish larvae.Using this property Hg2 + in zebrafish body distribution and toxicity of a preliminary study.
