目的探讨膀胱尿路上皮癌中水通道蛋白1、3(AQP1、AQP3)的表达与分布,及其与组织学分级和病理分期之间的关系及临床意义。方法采用免疫组化S-P法、蛋白免疫印迹(Western-blot)法对60例膀胱尿路上皮癌组织及30例正常膀胱组织中AQP1及AQP3的表达进行检测,并分析与其不同组织学分级、不同病理分期和有无淋巴结转移的关系。结果 AQP1在膀胱尿路上皮癌组织及正常膀胱组织中主要表达在微血管的内皮细胞的细胞膜及细胞质中,AQP3主要表达在膀胱尿路上皮癌的癌细胞的胞膜及胞质中。免疫组织化学S-P法和Western blot法检测的结果基本一致。正常膀胱组织与不同分级的膀胱尿路上皮癌组织中AQP1及AQP3的蛋白表达量分别比较,差异均有统计学意义(P<0.01);正常膀胱组织与不同病理分期的膀胱尿路上皮癌组织中AQP1及AQP3的蛋白表达量比较,差异均有统计学意义(P<0.01);AQP1及AQP3在有淋巴结转移的膀胱癌组织中的表达量分别与无淋巴结转移的膀胱癌组织比较,差异均有统计学意义(P<0.01)。结论 AQP1和AQP3的表达与膀胱癌的组织学分级及病理分期有关,在膀胱癌形成及发展过程中起重要作用,对预测膀胱癌的侵袭性、分期及临床诊疗有指导意义。
Objective To investigate the expression and distribution of AQP1 and AQP3 in bladder urothelial carcinoma and its relationship with histological grade and pathological stage and its clinical significance. Methods The expressions of AQP1 and AQP3 in 60 cases of bladder urothelial carcinoma and 30 cases of normal bladder tissues were detected by immunohistochemical SP method and Western-blot method. The expression of AQP1 and AQP3 in different tissues were analyzed with different histological grade and different Relationship between pathological stage and lymph node metastasis. Results AQP1 was mainly expressed in the plasma membrane and cytoplasm of endothelial cells of the microvessels in bladder urothelial carcinoma tissues and normal bladder tissues. AQP3 was mainly expressed in the membrane and cytoplasm of cancer cells in bladder urothelial carcinoma. Immunohistochemistry S-P method and Western blot test results are basically the same. The protein expressions of AQP1 and AQP3 in normal bladder tissues and different grades of bladder urothelial carcinoma tissues were significantly different (P <0.01), while those in normal bladder tissues and bladder cancer tissues (P <0.01). The expression levels of AQP1 and AQP3 in bladder cancer with lymph node metastasis were respectively compared with those without lymph node metastasis, the differences were statistically significant There was statistical significance (P <0.01). Conclusion The expression of AQP1 and AQP3 is related to the histological grade and pathological stage of bladder cancer and plays an important role in the formation and development of bladder cancer. It is of guiding significance in predicting the invasion, staging and clinical diagnosis and treatment of bladder cancer.
