目的:研究二乙基二硫代氨基甲酸钠(Sodiumdiethyldithiocarbamate,DTC)与狗、大鼠、小鼠、兔和人血浆的蛋白质结合率及DTC与后两者血浆蛋白的结合参数。方法:蛋白结合率用平衡透析法测定,DTC浓度用柱切换高效液相色谱法测定。结果:DTC血浆浓度在200~400umol·L-1时,DTC与狗、大鼠、小鼠和兔血浆蛋白质结合率分别高达(87.31±0.23)%,(89.91±0.17)%,(90,12±0.24)%和(89.96±0.31)%,而人的血浆蛋白质结合率仅为(68.63±0.55)%。DTC与兔和人血浆蛋白结合参数:β分别为8.54,0.29umol·g-1,kdp分别为72.91,5.49umol·L-1,K分别为16.82,135.45L·mmol-1。DTC与兔、人血浆蛋白结合部位数(N)分别为0.99,0.042。结论:DTC与兔、人血浆蛋白均具单一类型的结合部位,与人血浆蛋白结合率低于其他受试动物的血浆蛋白。
OBJECTIVE: To study the protein binding rate of sodium diethyldithiocarbamate (DTC) in dogs, rats, mice, rabbits and human plasma and the binding parameters between DTC and the latter two plasma proteins. Methods: The protein binding rate was determined by equilibrium dialysis, and the DTC concentration was determined by column switching high performance liquid chromatography. RESULTS: The plasma protein binding rates of DTC to dogs, rats, mice and rabbits were as high as (87.31 ± 0.23)% and (89.91 ± 0.23)%, respectively, at DTC plasma concentrations of 200-400 μmol·L-1. 17%, (90,12 ± 0.24)% and (89.96 ± 0.31)% respectively, while the plasma protein binding rate of human was only (68.63 ± 0.55)%. DTC and rabbit plasma protein binding parameters: β were 8.54,0.29umol · g-1, kdp were 72.91,5.49umol·L-1, K were 16.82,135.45L · Mmol-1. DTC and rabbit, human plasma protein binding sites (N) were 0.99,0.042. CONCLUSION: Both DTC and rabbit and human plasma proteins have a single type of binding site with a lower binding rate to human plasma proteins than plasma proteins of other test animals.
