通过观察系统性呼吸道变态反应模型小鼠脾脏T辅助淋巴细胞的变化,探讨Th1/Th2失衡以及玉屏风散对其的影响。30只Balb/c小鼠按随机数字表法均分为正常对照组、玉屏风散治疗组和模型组。建立用OVA刺激激发的上下呼吸道变态反应性炎症模型,采用流式细胞术分别测定CD3+、CD4+以及细胞内IFNγ和IL4,治疗组在激发同时给予玉屏风散腹腔注射(2000mg/kg,7d)。结果显示,系统激发动物模型组显示明显非特异性炎症反应征象,激发同时给予中药玉屏风散组可显著改善炎症反应征象。治疗组Th1和Th1/Th2值显著高于正常组和模型组(P<0.01)。治疗组和模型组Th2显著高于正常组(P<0.01),而治疗组与模型组之间无显著差异。提示玉屏风散对小鼠系统性呼吸道变态反应模型在激发期干预治疗作用明显,上调Th1/Th2比值可能是其药效作用机制之一。
By observing the change of T helper lymphocyte in spleen of mice with systemic respiratory allergic reaction, the imbalance of Th1/Th2 and the influence of Yupingfengsan on it were discussed. Thirty Balb/c mice were divided into normal control group, Yupingfeng Powder treatment group and model group according to the random number table method. The allergic inflammation models of upper and lower respiratory tract stimulated by OVA stimulation were established. CD3+, CD4+ and intracellular IFNγ and IL4 were measured by flow cytometry. The treatment group was given intraperitoneal injection of YP (2000 mg/kg, 7 d). The results showed that the system stimulated the animal model group to show obvious signs of non-specific inflammatory reaction, and the simultaneous administration of the Chinese medicine Yupingfeng San group could significantly improve the signs of inflammation. The Th1 and Th1/Th2 values in the treatment group were significantly higher than those in the normal group and the model group (P<0.01). Th2 was significantly higher in the treatment group and the model group than in the normal group (P<0.01), but there was no significant difference between the treatment group and the model group. It is suggested that the systemic respiratory allergic reaction model of mice treated by Yupingfeng Powder plays an important role in interventional therapy during the provocation period, and up-regulation of Th1/Th2 ratio may be one of the mechanisms of its efficacy.
