目的 观察高糖状态下肾小球系膜细胞 (GMC)增殖及纤维连接蛋白 (FN)合成情况。 方法 采用体外 GMC培养 ,噻唑蓝 (MTT)掺入法和细胞计数观察 GMC增殖 ,用 RT— PCR及双抗夹心 EL ISA方法检测 FN的基因及蛋白质水平变化。 结果 高糖对 GMC增殖有抑制作用 ,对 FN的合成呈浓度及作用时间依赖性增高。结论 高血糖在糖尿病肾病的发生发展过程中起重要作用。
Objective To observe the proliferation of glomerular mesangial cells (GMCs) and the synthesis of fibronectin (FN) under high glucose conditions. Methods GMC proliferation, MTT incorporation and cell counting were used to observe the proliferation of GMC. The changes of FN gene and protein levels were detected by RT-PCR and double-antibody sandwich ELISA. Results High glucose inhibited the proliferation of GMC, and the concentration of FN and the time-dependent effect were increased. Conclusion Hyperglycemia plays an important role in the development of diabetic nephropathy.
