采用荧光分光光度法 ,在体内、体外研究了溴氰菊酯对大鼠脑组织中EROD( 7 ethoxyresorufinO deethylase)酶活性的影响。结果表明 :大鼠经溴氰菊酯 12 5mg·kg-1·d-1通过腹腔注射连续 5天后 ,溴氰菊酯明显抑制全脑和大脑皮层EROD酶活性 ,抑制率分别是 3 2 0 % ,3 7 0 % ;在体外 ,溴氰菊酯浓度在 2× 10 -8~ 2× 10 -4 mol/L范围内对EROD活性无明显影响 ;进一步用蛋白印迹技术检测CYP1A1/1A2蛋白表达的变化 ,结果表明 :溴氰菊酯在体内明显抑制上述各脑区中CYP1A1/1A2蛋白表达。综合上述两方面结果 ,提示溴氰菊酯在体内能明显抑制脑内EROD活性 ,其机理与抑制该酶蛋白合成有关。
Fluorescence spectrophotometry was used to study the effect of deltamethrin on the enzyme activity of EROD (7 ethoxyresorufinO deethylase) in rat brain in vitro and in vivo. The results showed that deltamethrin significantly inhibited the activity of EROD in whole brain and cerebral cortex after 5 mg · kg-1 · d-1 deltamethrin injection for 5 consecutive days in rats. The inhibitory rates were 30.2% , While the concentration of deltamethrin in the range of 2 × 10 -8 ~ 2 × 10 -4 mol / L had no significant effect on the activity of EROD in vitro. Western blotting was used to detect the expression of CYP1A1 / 1A2 protein The results showed that deltamethrin significantly inhibited CYP1A1 / 1A2 protein expression in the above brain regions in vivo. Based on the above two results, it is suggested that deltamethrin can significantly inhibit brain EROD activity in vivo, and its mechanism is related to the inhibition of the synthesis of the enzyme protein.
