本文用3H-胸腺嘧啶脱氧核苷掺入法研究了八肽胆囊收缩素(CCK8)及其受体拮抗剂对人胰腺癌细胞株(PU-PAN-1)细胞生长的作用,并观察了CCK8对癌细胞膜和胞浆中蛋白激酶C(PKC)活性的影响。结果表明,CCK8可显著刺激人胰腺癌细胞的生长(35%~50%),而此促生长作用可被CCK受体拮抗剂所抑制。CCK8也可使癌细胞膜和胞浆中PKC活性分别升高2~16倍和5~14倍。CCK8刺激细胞生长和PKC活性增高的浓度是一致的,这提示PKC可能介导CCK8对人胰腺癌细胞的促生长作用。
In this paper, the effect of CCK8 and its receptor antagonist on the growth of human pancreatic cancer cell line PU-PAN-1 was studied by 3H-thymidine incorporation, and CCK8 was observed. Effects on the activity of protein kinase C (PKC) in cancer cell membranes and cytoplasm. The results show that CCK8 can significantly stimulate the growth of human pancreatic cancer cells (35% to 50%), and this growth-promoting effect can be inhibited by CCK receptor antagonists. CCK8 also increased the PKC activity in cancer cell membranes and cytoplasm by 2 to 16 fold and 5 to 14 fold, respectively. The concentration of CCK8-stimulated cell growth and increased PKC activity is consistent, suggesting that PKC may mediate CCG8’s promoting effect on human pancreatic cancer cells.
