目的初步探讨HAART治疗后CD4+T细胞的CD44、CD95分子的表达水平变化。方法采用流式细胞仪检测CD4+处女型T细胞(CD45RA+T)和记忆型(CD45RO+T)表面CD44、CD95的表达水平,并分析二者的相关性。结果HIV-1感染组CD4+处女型T细胞(CD45RA+T)和记忆型T细胞(CD45RO+T)表面CD44分子的表达分别为21.17±8.36和28.40±10.49,比正常对照组的37.65±2.92和51.03±4.12低,具有显著性差异;与CD44分子的表达相反,HIV-1感染组CD4+处女型T细胞(CD45RA+T)和记忆型T细胞(CD45RO+T)表面CD95分子的表达分别为2.54±1.49和19.19±6.96,比正常对照组的1.51±0.11和13.68±1.37高,具有显著性差异。按照美国CDC对于艾滋病的临床分期标准对HIV感染者进一步分期,然后进行比较后,总体上仍反映出上述特点。另外,CD44和CD95的异常表达在B期尤为明显。经过HAART治疗以后,上述特点和异常能在一定程度上得到改变,但不能恢复,与对照组之间仍然具有显著性差异。研究还发现,细胞表面CD95和CD44的表达具有显著的正相关性。结论CD44分子的水平并不能代表体内病毒的拷贝数。HAART治疗后,新生的细胞在向淋巴结归巢的过程中,细胞凋亡增加。
Objective To investigate the expression of CD44 and CD95 in CD4 + T cells after HAART treatment. Methods The expression of CD44 and CD95 on CD4 + virion T (CD45RA + T) and memory (CD45RO + T) T cells was detected by flow cytometry. The correlation between them was analyzed. Results The expression of CD44 on CD4 + T cells (CD45RA + T) and memory T cells (CD45RO + T) in HIV-1 infection group was 21.17 ± 8.36 and 28.40 ± 10.49 respectively, which was significantly higher than that in the normal control group (37.65 ± 2.92 and 51.03 ± 4.12, respectively. Compared with the expression of CD44, the expression of CD95 on CD4 + virgin T cells (CD45RA + T) and memory type T cells (CD45RO + T) in HIV-1 infected group was 2.54 ± 1.49 and 19.19 ± 6.96, respectively, which were significantly higher than those of the control group (1.51 ± 0.11 and 13.68 ± 1.37). According to the United States CDC for clinical staging of HIV in HIV-infected patients further staging, and then compared to the overall still reflect the above characteristics. In addition, aberrant expression of CD44 and CD95 is particularly evident in stage B. After the treatment of HAART, the above characteristics and abnormalities can be changed to some extent, but can not be recovered, and there is still a significant difference between the control group. The study also found that the cell surface expression of CD95 and CD44 has a significant positive correlation. Conclusion The level of CD44 does not represent the copy number of virus in vivo. After HAART treatment, newborn cells increase their apoptosis during homing to the lymph nodes.
