目的研究阿司匹林对醛固酮诱导心肌成纤维细胞(CFs)增殖的抑制作用。方法将培养3~5代SD新生大鼠的CFs分为空白组、模型组、阿司匹林组(1.11μmol·L-1)和阳性组(螺内酯1μmol·L-1);用Giemsa染色法观察各组CFs的形态学变化,用ELISA法分别检测Ⅰ、Ⅲ型胶原纤维,采用比色法分析上清液中羟脯氨酸的含量。结果阿司匹林可有效抑制醛固酮诱导的CFs增殖;还可显著抑制醛固酮诱导的CFs中Ⅰ、Ⅲ型胶原纤维的分泌。结论阿司匹林对CFs的增殖及胶原分泌有一定的抑制作用,具潜在的抗心肌纤维化作用。
Objective To study the inhibitory effect of aspirin on aldosterone-induced proliferation of cardiac fibroblasts (CFs). Methods CFs cultured in neonatal SD rats of 3 to 5 generations were divided into blank group, model group, aspirin group (1.11 μmol·L-1) and positive group (spironolactone 1 μmol·L-1). The Giemsa staining The morphological changes of CFs were detected by ELISA method Ⅰ, Ⅲ collagen fibers, the colorimetric analysis of hydroxyproline content in the supernatant. Results Aspirin can effectively inhibit the aldosterone-induced CFs proliferation, but also significantly inhibited aldosterone-induced CFs in type Ⅰ, Ⅲ collagen fibers. Conclusions Aspirin can inhibit the proliferation and collagen secretion of CFs to a certain extent, with potential anti-myocardial fibrosis effect.
