目的通过构建traP突变菌株研究RNAⅢ激活蛋白靶蛋白(TRAP)蛋白在金黄色葡萄球菌(SAU)中的功能。方法利用同源重组的方法在金黄色葡萄球菌(SAU)8325-4中构建traP的缺失突变菌株,对其表型和几种压力条件下的生长和生存状况进行检测,揭示其在金黄色葡萄球菌生命活动中的作用。结果在金黄色葡萄球菌8325-4中成功构建了traP的缺失突变菌株,突变株生长缓慢,其他表型如疏水性、表面电荷和生物膜发生了明显的变化;在重金属、过氧化氢,以及全血杀伤等压力条件下的生长和生存状况也发生了明显改变。结论TRAP蛋白在金黄色葡萄球菌侵袭机体的过程中发挥着重要的功能。
OBJECTIVE: To study the function of RNA Ⅲ activating protein target protein (TRAP) in Staphylococcus aureus (SAU) by constructing traP mutants. Methods The homologous recombination method was used to construct the deletion mutant of trap in Staphylococcus aureus (SAU) 8325-4. The phenotypes and the growth and survival status under several stress conditions were detected, The role of cocci in the life activity. Results The mutant strains of traP were successfully constructed in Staphylococcus aureus 8325-4. The mutants grew slowly. Other phenotypes such as hydrophobicity, surface charge and biofilm changed significantly. In the heavy metals, hydrogen peroxide and The growth and survival conditions under whole blood kill and other stress conditions have also undergone significant changes. Conclusion TRAP protein plays an important role in the invasion of Staphylococcus aureus in the body.
