目的:研究肾衰诱发心衰大鼠心脏钙泵活性及基因表达的变化。方法:雄性SD大鼠行5/6肾切除,6周后造成慢性肾衰,8周后诱发左室心衰。Bradford法测定24h尿蛋白量,试剂盒测定血清肌酐尿素氮。全导联测定大鼠心电图。左心室插管测定心脏血流动力学指标。称重法测定大鼠体重及心脏重量参数。测定大鼠心脏钠钾泵、钙泵活性及相关基因的表达。结果:血生化与尿蛋白指标指示慢性肾衰造模成功,血流动力学及心电图心指数等指标表明大鼠已诱发左心心衰。模型组大鼠心肌质膜钠钾泵及肌浆网钙泵活性均较假手术组大鼠降低且有统计学意义,荧光定量PCR测定相关基因的表达也明显减少。结论:慢性肾衰诱发心衰主要由于影响心脏钙泵及钠钾泵基因表达,相关酶的活性受到影响,损伤程度随时间延长而加深。
Objective: To study the changes of cardiac calcium pump activity and gene expression in heart failure rats with renal failure. Methods: Male SD rats were subjected to 5/6 nephrectomy. Chronic renal failure was induced after 6 weeks and left ventricular heart failure was induced after 8 weeks. The amount of urinary protein was measured by Bradford method and serum creatinine was measured by kit. All lead determination of rat electrocardiogram. Left ventricular intubation for the determination of cardiac hemodynamic parameters. Weighing method was used to determine body weight and heart weight parameters. The sodium potassium pump, calcium pump activity and expression of related genes in rat heart were determined. Results: Blood biochemistry and urinary protein index of chronic renal failure model success, hemodynamics and ECG index and other indicators show that rats have been induced left heart failure. The Na + -K + pump and sarcoplasmic reticulum calcium pump activity in the model group were significantly lower than those in the sham operation group, and the expression of related genes in the model group was also significantly decreased. CONCLUSION: Chronic renal failure induced by heart failure mainly affects the expression of cardiac calcium pump and sodium-potassium pump, the activity of related enzymes is affected, and the degree of injury deepens with time.
