目的:探讨瘤内125I粒子植入对Lewis肺癌细胞生长及凋亡相关蛋白Caspase-9、Caspase-3表达的影响。方法:昆明小鼠被注入1×107个瘤细胞(0.2ml),当肿瘤生长至0.20-2.5cm2大小时,随机分为3组(对照组、5Gy组和10Gy组),分别植入粒子。观察并测量肿瘤的生长曲线和抑瘤率,应用免疫组织化学技术检测125I粒子照射后肿瘤凋亡相关蛋白Caspase-9、Caspase-3的表达。结果:粒子植入前各组平均瘤体积差异无统计学意义(p>0.05),但是植入后1.0mCi组在第4天(5Gy)和第9天(10Gy)时远小于对照组;0.5mCi组在第9天(5Gy)时瘤体变化不大,在第19天(10Gy)时小于对照组(p>0.05)。同时Caspase-9和Caspase-3的表达基本与肿瘤体积变化一致(p<0.05orp<0.01)。结论:125I粒子低剂量照射能够明显的控制肿瘤生长并可促使凋亡相关蛋白Caspase-9、Caspase-3高表达,使肿瘤细胞凋亡,最终杀灭肿瘤。这可能是粒子照射诱导细胞凋亡从而杀伤肿瘤的途径之一。
OBJECTIVE: To investigate the effect of intratumoral 125I implantation on the growth of Lewis lung carcinoma cells and the expression of Caspase-9 and Caspase-3. Methods: Kunming mice were injected with 1 × 107 tumor cells (0.2ml). When tumors grew to 0.20-2.5cm2, they were randomly divided into 3 groups (control group, 5Gy group and 10Gy group). The growth curve and tumor inhibition rate were observed and measured. The expression of Caspase-9, Caspase-3 and tumor necrosis factor-related protein (IAP) were detected by immunohistochemistry. Results: There was no significant difference in mean tumor volume between groups before implantation (p> 0.05), but 1.0mCi group was much smaller than control group on day 4 (5Gy) and day 9 (10Gy) The tumors in the mCi group did not change much at day 9 (5 Gy) and were smaller at day 19 (10 Gy) than in controls (p> 0.05). At the same time, the expression of Caspase-9 and Caspase-3 was basically the same as the change of tumor volume (p <0.05orp <0.01). Conclusion: Low doses of 125I can significantly inhibit the tumor growth and induce the expression of Caspase-9 and Caspase-3, which can induce the apoptosis of tumor cells and eventually kill the tumor. This may be one of the ways that particle irradiation induces apoptosis and thus kills the tumor.
