目的 :了解菘蓝 4种有效单体 (6 ,7,8,9号 )抗柯萨奇病毒B组 3型 (CVB3 )的活性。方法 :通过观察细胞病变效应 (CPE) ,噻唑蓝 (MTT)比色法检测细胞活性 ,用半定量RT PCR检测不同药物浓度下病毒核酸含量 ,以确定有效单体抗CVB3 的作用。结果 :菘蓝的 4种单体成分对CVB3 生物合成均有抑制作用 ,且强于阳性对照药物病毒唑。表现在随着药物浓度的增加 ,CPE程度逐渐减弱 ,病毒核酸的含量逐渐下降。结论 :菘蓝的 4种单体能在体外通过抑制生物合成发挥抗CVB3 作用。
AIM : To understand the activity of four effective monomers (6, 7, 8, 9) against Coxsackie virus B group 3 (CVB3) in indigo. Methods: Cell viability was measured by cytopathic effect (CPE) and thiazolyl blue (MTT) colorimetric assay. Semi-quantitative RT-PCR was used to determine the concentration of viral nucleic acid at different drug concentrations to determine the effect of effective monomeric anti-CVB3. Results : The four monomeric components of indigo inhibited the biosynthesis of CVB3 and were stronger than the positive control ribavirin. With the increase of drug concentration, the degree of CPE gradually weakens, and the content of viral nucleic acid gradually decreases. Conclusion : The four monomers of indigo can exert anti-CVB3 activity through inhibiting biosynthesis in vitro.
