目的:考察姜蚬护肝汤对酒精性肝损伤小鼠的保护作用及其机制。方法:小鼠按体重随机为正常组,模型组,姜蚬护肝汤低、中、高剂量组,每组10只,连续灌胃给药30 d后,灌胃50%乙醇溶液14 ml·kg~(-1),禁食16h后取血,取肝脏;测定血清中的TNF-α、AST、ALT的水平;测定肝组织中MDA、GSH、TG、Caspase-3、Caspase-8和NF-κB的水平;HE染色后观察肝组织病理学改变。结果:姜蚬护肝汤高剂量组小鼠的TNF-α、AST、ALT、MDA、TG、Caspase-3、Caspase-8、NF-κB均较模型组显著降低,GSH则显著提高(P<0.05);肝脏脂肪变性及组织坏死情况有所减轻。结论:姜蚬护肝汤对实验性酒精性肝损伤具有良好的对抗作用,其可能作用机制为降低TNF-α和NF-κB的过度表达从而减轻肝细胞的炎症反应,并减少Caspase-3、Caspase-8诱导的肝细胞凋亡。
Objective: To investigate the protective effect and mechanism of Ganyu Hugan Decoction on alcoholic liver injury in mice. Methods: The mice were randomly divided into normal group, model group, Jiangpi Hugan Decoction low, medium and high dose group, 10 rats in each group. After continuous intragastric administration for 30 days, the mice were treated with 50% ethanol solution The levels of TNF-α, AST and ALT in the serum were measured. The levels of MDA, GSH, TG, Caspase-3, Caspase-8 and NF -κB levels; HE staining liver pathological changes. Results: The levels of TNF-α, AST, ALT, MDA, TG, Caspase-3, Caspase-8 and NF-κB in the Jiang dose Hugan decoction high dose group were significantly lower than those in the model group 0.05); liver steatosis and tissue necrosis have eased. CONCLUSION: Jianggan Hugan Decoction has a good antagonistic effect on experimental alcoholic liver injury. Its possible mechanism is to reduce the over-expression of TNF-α and NF-κB, thereby reducing the inflammatory reaction of hepatocytes and decreasing the expression of Caspase-3, Caspase-8-induced hepatocyte apoptosis.
